Free intracellular Ca was measured in a cultured Purkinje neuron using fura-2 microfluorimetric imaging. The neuron in this picture was bathed in Ca-free external saline and was responding to application of trans-ACPD, an agonist of metabotropic glutamate receptors. In this image, blue pixels represent a Ca concentration of about 50 nM and red pixels, about 500 nM. Activation of the mGluR1 subtype of this receptor is an important event in triggering cerebellar long-term depression, a possible cellular substrate of memory.
