Neurobiology of Degenerative Brain Diseases in Animals and Man

     Investigations in the Neuropathology Laboratory focus on: clinical-pathological correlations; studies of the neuronal cytoskeleton toxicity of mutant proteins in experimental models and human illnesses, including Alzheimer’s Disease (AD), Parkinson's Disease, and other neurodegenerative diseases, Amyotrophic Lateral Sclerosis (ALS) and nerve injury/regeneration; the mechanisms leading to selective dysfunction/death of populations of neurons in clinical and experimental neurological diseases; analyses of transgenic mouse models expressing mutant genes linked to AD (ie. amyloid precursor protein and presenilin 1 [PS1] genes), to familial ALS (superoxide dismutase 1 [SOD1]), and to familial PD (synuclein); biological and biochemical studies of the processes whereby mutations cause neuronal abnormalities in vitro and in vivo; analyses of the outcomes of targeting specific gene (e.g. PS1, BACE1, nicastrin, CCS, etc) to asses the impact of specific gene products on the biochemical/neuropathological abnormalities in mouse models; and experimental therapies (including gene targeting studies that are used to develop and validate new therapeutic targets) in animal models of neurodegenerative disease.
